Se ha observado que el D-003 (5 y 25 mg/kg) previene el daño hepático inducido por paracetamol,
ya que reduce significativamente el área necrótica y los infiltrados inflamatorios en este tejido.
Considerando que el incremento del estrés oxidativo es el principal mecanismo del daño hepático inducido
por paracetamol y considerando los efectos antioxidantes del D-003, este estudio consistió en investigar si
el tratamiento oral con D-003, administrado a dosis en que previene del daño en hígado contra el paracetamol,
pudiera modificar favorablemente parámetros asociados al estrés oxidativo. La administración oral
de D-003 (5-200 mg/kg) durante 4 semanas disminuyó, de manera dosis dependiente, el incremento de las
enzimas plasmáticas aspartato aminotransferasa (ASAT), alaninoaminotransferasa (ALAT) y lactato
dehidrogenasa (LDH) inducido por la administración de paracetamol (650 mg/kg) en ratas. El efecto máximo
se logran con 50 mg/kg. Además el D-003 reduce significativamente y de manera dosis dependiente el
incremento inducido por el paracetamol sobre las concentraciones de hidroperóxidos totales (HT) y malondialdehido
(MDA) en tejido hepático. Así, el D-003 (50 mg/kg) restituyó los niveles de grupos tioles (SH)
e incrementó significativamente la actividad de la catalasa (CAT) y la glutatión peroxidasa (GPX), las cuales
fueron disminuidas por el paracetamol. Estos resultados son consistentes con datos previos que plantea
que el D003 inhibe la peroxidación lipídica (PL) en modelos experimentales y humanos, y se ha descrito
que el D003 protege sobre el daño hepático inducido por tetracloruro de carbono en que la LP exhibe el rol
principal. Además, el tratamiento con D-003 también aumento la actividad de la GPX y la glutation reductasa
(GSHR) Así, el hecho de que el D-003 incremente la actividad de las enzimas, sugiere un mecanismo
protector adicional del D-003 que puede manifestarse en condiciones extremas de desequilibrio oxidativo. D-003 (5 and 25 mg/kg) has shown to prevent paracetamol-

that increased oxidative stress is a main mechanism of paracetamol-induced liver damage and the antioxidant
effects of D-003, this study was undertaken to investigate whether oral treatment with D-003, administered at
doses able to prevent paracetamol liver damage could favorably modify parameters associated to oxidative stress.
D-003 (5-200 mg/kg) orally administered for 4 weeks significantly reduced, in a dose dependent manner, the increases
of plasma enzymes aspartate aminotransferase (AST), alaninoaminotransferase (ALT) and lactate dehidrogenase
(LDH) induced with paracetamol (650 mg/kg) in rats, the maximal effects being achieved with 50
mg/kg. Also, D-003 significantly and dose dependently reduced the increases of total hydroperoxides (HT) and
malondialdehyde (MDA) liver concentrations induced with paracetamol. D-003 (50 mg/kg) contributed to restore
the levels of thiol groups (SH), as well as the catalase (CAT) and glutathione peroxidase (GPX) activities,
all decreased with paracetamol. These results are consisting with previous data supporting that D-003 inhibits
lipid peroxidation (PL) in experimental models and humans, and with the protective effects described for D-003
on the liver damage induced with CCl4 in which LP displays the pivotal role. Moreover, since D-003 increased
the activity GPX and GSH-reductase (GSHR) decreased with paracetamol suggest and additional mechanism
probably present in extreme conditions of unbalanced oxidative cell status. |
